Strain Comments:

• gyrA42(ts)-- has a high spontaneous reversion frequency. We suggest frequent verification of the Ts phenotype.
• thyA45-- was called tlr-25 and thyR25, in conflict with the thyA25 allele. The Tlr (Thy low req.) phenotype is now known to result from two mutations, thyA^--, deo^-.
• thyA45-- Strains with this marker must have their medium supplemented with thymine (25ug/ml). This is true even for rich medium such as LB. Otherwise thymine starvation can lead to thymineless death.
• polA0-- The source of this allele is not yet known.
• deo-225-- is in deoB or deoC. In combination with a thyA^- allele, it results in low Thy-requirement (Tlr) phenotype.
• deo-225 was formerly called thyA45
• deo-225 was formerly called thyR25
• deo-225 was formerly called tlr-25
• ts = Temperature Sensitive
